Forebrain Acetylcholine Modulates Isoflurane and Ketamine Anesthesia in Adult Mice

چکیده

Background Cholinergic drugs are known to modulate general anesthesia, but anesthesia responses in acetylcholine-deficient mice have not been studied. It was hypothesized that with genetic deficiency of forebrain acetylcholine show increased anesthetic sensitivity isoflurane and ketamine decreased gamma-frequency brain activity. Methods Male adult heterozygous knockdown vesicular transporter the or homozygous knockout basal were compared wild-type mice. Hippocampal frontal cortical electrographic activity righting reflex studied response doses. Results The loss-of-righting-reflex dose for lower (mean ± SD, 0.76 0.08%, n = 18, P 0.005) (0.78 0.07%, 24, 0.021), as (0.83 23), using a significance criterion 0.017 three planned comparisons. Loss-of-righting-reflex (144 39 mg/kg, 14, 0.006) (162 32 20, 0.602) (168 24 21). high-gamma (63 100 Hz) power after significantly (isoflurane-dose mouse-group interaction effect, F[8,56] 2.87, 0.010; 5 6 per group). both when (interaction effect F[2,13] 6.06, 0.014). change gamma statistically different among knockout, knockdown, Conclusions These findings suggest cholinergic neurons behavioral hippocampal during anesthesia. Basal forebrain histaminergic transmission modulates electroencephalographic activity and emergence from isoflurane anesthesia.

BACKGROUND The tuberomammillary histaminergic neurons are involved in the sedative component of anesthetic action. The nucleus basalis magnocellularis (NBM) in the basal forebrain receives dense excitatory innervation from the tuberomammillary nucleus and is recognized as an important site of sleep-wake regulation. This study investigated whether NBM administration of histaminergic drugs may mo...

Successful Small Intestine Colonization of Adult Mice by Vibrio cholerae Requires Ketamine Anesthesia and Accessory Toxins

Vibrio cholerae colonizes the small intestine of adult C57BL/6 mice. In this study, the physical and genetic parameters that facilitate this colonization were investigated. Successful colonization was found to depend upon anesthesia with ketamine-xylazine and neutralization of stomach acid with sodium bicarbonate, but not streptomycin treatment. A variety of common mouse strains were colonized ...

The effect of isoflurane anesthesia on memory

Memory and learning are considered as the most complicated and important processes of behavioral sciences. On the other hand, considering the wide .and progressively increasing applications of anesthetic agents all over the world, it is of great importance to have a reasonable knowledge regarding their effects on the memory process and its retrieval. The main purpose of this study was to evalua...

Ketamine: use in anesthesia.

The role of ketamine anesthesia in the prehospital, emergency department and operating theater settings is not well defined. A nonsystematic review of ketamine was performed by authors from Australia, Europe, and North America. Results were discussed among authors and the final manuscript accepted. Ketamine is a useful agent for induction of anesthesia, procedural sedation, and analgesia. Its p...
